Comparing the phenotype of PHPIa patients harboring deleterious truncating mutations (only nonsense and frameshift mutations, which most probably cause a loss of protein activity, n = 33), with those patients harboring missense mutations ( n = 31), we observed a highly significant difference on subcutaneous calcifications ( P = 2.6e-06).
bottom 1000 associated transcripts for each of the 61 and 43 trans-SNPs, respectively, we find a highly significant enrichment for lower P-values in the top 1000 transcripts (Mann-Whitney, ALSPAC P = 2.6 × 10 −6 and Oxford P=3.8 × 10 −5 ).
In fact, when we compared the mean firing rate evoked by light activation of all these presumably ‘non-inhibitory’ spots to the mean baseline activity, we found that there was a small but highly significant reduction in firing rate (P = 2.6e-6, paired t-test, n = 216 spots, spot size 88 × 88 mμ, each spot value is the represented by the average of its ~20 repetitions, Fig. 2B ).
Although the remaining histological patterns were similar between ERD and NERD in HH+ (Figure 3 ), basal zone hyperplasia and loss of glycogen accompanied HH in all cases, with the correlation being highly significant in NERD ( P = 2.61 × 10 -6 ) (Figure 4 ).

The Pearson test also showed a highly significant negative correlation between the presence of symptoms and QoL with a correlation coefficient = -0.47 and p-value = 2.63 × 10^6 (Fig. 1 ).
The upregulation of G6PD and PGLS in the PR Low divergent group was highly significant ( p = 2.63E-06 and p = 9.78E-05), and PGD and RPIA also showed a clear trend of increased expression ( p = 0.0939 and p = 0.1170), contributing to a more pronounced overall activation pattern of the pathway (Fig.
In contrast, the difference in the overall severities of disease when E7 was turned off in the FancD2 -sufficient background was highly significant (for Bi-L E7 / K14 – tTA / FancD2 +/+ versus Bi-L E7 / K14 – tTA / FancD2 +/+ [Dox], P = 2.644 × 10 −6 ; P = 7.9 × 10 −6 if we exclude cancers).
Validation of genetic variants Validation of all nine potentially deleterious genetic variants detected in whole genome sequencing data in 102 SPAID-affected and 62 SPAID-unaffected Shar-Pei using Kompetitive Allele Specific PCR (KASP) and gel electrophoresis revealed a borderline significant P-value for TGFBR3 :g.57204844A > G located in TGFBR3 (transforming growth factor beta receptor 3; P = 0.050), but a highly significant P-value for MTBP :g.19383758G > A (P = 2.664E-06) located in MTBP (Mdm2, transformed 3 T3 cell double minute 2, p53 binding protein ).
SLC39A8 and the central nervous system Another early GWAS discovery of a SLC39A8 variant involved a case-control study of 476 schizophrenia patients and 447 control subjects from Galicia, combined with a replication sample comprising >4000 cases and >15,100 control subjects of European origin ; the SNV (rs13107325; p.Ala391Thr) was somewhat significant ( P = 2.7 × 10 −6 ) in the collective sample, following Bonferroni correction [ 60 ].
This yielded a highly significant association of p = 2.71 × 10 −6 and OR = 3.15 (95% CI: 1.95, 5.09) for the heterozygous genotype under an additive model.
However, FIMO analysis detected a highly significant motif sequence, GGGAGGCTA ( p = 2.72 × 10−6; q = 9.77 × 10−4), which was used as the representative fragment for docking (Fig. 7 B).
In CGG 4, 10 of the 14 transcripts are annotated [six are ribosomal proteins ( Rps29 , Rps15 , Rplp2 , Rplp1 , Rpl35A , and Rpl19 ), and two are ribosomal protein/ubiquitin fusions ( Fau and Uba52 )] and showed a highly significant enrichment for translation (GO:0006412; P = 2.77 × 10 −6 ).
The average number of trichomes per square millimeter of stem surface in the OE lines was 306, representing a highly significant increase of 13.40% compared to the WT (265 trichomes per square millimeter, p = 2.78 × 10 −6 ).
Notably, 8 of these 12 genes ( ENO2, LUC7I3, KIF21A, MAP4K4 , PPP3CA, MACF1, NDRG4, and PRRC2C ) were differentially spliced in 12-month-old Trem2*R47H mice, which was highly significant (hypergeometric p = 2.78 × 10 –6 ) (Table 3 ).
This difference was highly significant (Wilcoxon rank-sum test, false discovery rate (FDR)-adjusted P = 2.8 × 10 −6 ; Extended Data Fig. 4c–e and Supplementary Table 16 ).
As can be observed in Table 2 , the 10% DMSO treatment showed highly significant differences compared to all other groups ( p -value = 0.0000028), corroborating its cytotoxic profile [ 95 ].
Indeed, 3 of its markers were highly significant: rs41575887 (127,928,903 bp, p = 2.8E-06), rs132708238 (127,934,719 bp, p = 6.6E-10), and rs136864039 (127,944,097 bp, p = 1.5E-06).
But as we predicted, our RD analysis showed that RD>0.02 was extremely significant ( p = 2.81e-06, 5.12e-05, and 1.53e-07 in overall, control, and case populations, respectively, Table 6 ).
A study tested the association of CYP24A1 polymorphism and coronary artery calcification CAC in three populations, the Amish Family Calcification Study (AFCS), the Genetic Epidemiology Network of Arteriopathy (GENOA), and the Penn Coronary Artery Calcification (PennCAC) cohorts found that rs2762939 C allele consistently showed association with reduced coronary artery calcification, with meta-analysis yielding a highly significant result ( p =2.9×10 − 6) [ 5 ].
Eight of 20 genes were nominally associated at p ≤ 0.05 (3 genes at Bonferroni-adjusted p ≤ 0.05, accounting for 20 tests), consistent with highly significant enrichment (exact binomial test p = 2.9 × 10 −6 ; Table S5 ).
In this case, despite a highly significant correlation (ρ = −0.83, adj. p ≤ 2.93 × 10 −6 ), the wide confidence interval [−0.96, −0.45] indicates uncertainty in the estimate. 2.3.
The SNP Gga_rs13606164, located at 2579.1 kb on GGA26, is 35.6 kb downstream of the thyroid stimulating hormone, beta ( TSHB ) gene, had highly significant association with the polydactyly (P < 2.98 × 10 -6 ).
