Though not statistically significant, we also found an interesting trend with respect to the risk of contralateral invasive events.
Excerpts
Notably, a numerical trend towards inferior outcomes was observed in highly immunogenic brain metastases characterized by combined adenosine pathway activation and high PD-L1 expression (adenosine high/PD-L1 high), which may indicate a clinically relevant high-risk phenotype and further underscore the unmet medical need for an expansion of immunotherapeutic strategies in brain metastases.
These findings, which provide contemporary data that confirm an increasing trend in the prevalence of cancer among delivery hospitalizations, also extend prior work conducted in the United States and other European countries.
A decreasing trend in HR-HPV prevalence was observed with increasing age, with the exception of the oldest age group, where a slight increase was noted.
Even after the stringent Bonferroni correction for multiple testing, the associations with bone disease, high-risk cytogenetics, and MAPK mutational status remained highly significant ( p adj < 0.001), supporting the notion that mutations are present throughout the natural history of plasma cell dyscrasias ( Table 4 ).
Patients with mutations in signaling pathway genes ( FLT3 , NRAS , KRAS , NF1 , PTPN11 , JAK / STAT , KIT , CSF3R ) showed a trend to shorter survival, particularly evident for those with RAS/RTK pathway signaling [ 155 ].
Subgroup analyses stratified by primary tumor origin showed a trend toward shorter overall survival in patients with higher Ki-67 expression across spinal metastases from prostate, lung and breast cancer; however, these differences were not statistically significant within individual tumor entities ( Figure 2 C).
Second, multiple pairwise comparisons were performed across several correlated dosimetric endpoints without formal multiplicity adjustment, so an increased risk of type I error cannot be excluded, particularly for borderline significant findings.
For instance, patients on an SERD (fulvestrant) showed a higher hazard ratio (HR = 2.302, p = 0.0678), and those with non-visceral metastases (HR = 2.008, p = 0.0696) also showed a trend toward poorer outcomes without statistical significance. 3.4.
Leukaemia: The excess of leukaemia in parents in our study who were assumed not to have a germline RB1 mutation has no obvious explanation and, though statistically highly significant, may be due to chance.
Alterations in DNA damage response pathways and apoptosis/immune regulatory mechanisms also showed a trend toward enrichment in SS, with the latter observed only in SS tumors (4% vs. 0%), albeit at low frequency.
A significant reduction in SALL1 mRNA was observed in grade 3 (poorly differentiated) tumors, whereas grade 1 (well differentiated) and 2 (moderately differentiated) showed a decreasing trend without statistical significance.
a clear trendgold
In the AML19 study, AML patients were randomized to receive Tludarabine, Cytarabine, Idarubicin, and G-CSF (FLAG-IDA) or DA (Daunorubicin/Cytarabine): relapse rate (RR) was significantly lower with FLAG-IDA than with DA (three-year RR 26% or 68%, respectively), and there was a clear trend of improved OS for patients with FLAG-IDA compared to DA (three-year OS 66% vs. 37%, respectively) [ 28 ].
Overall survival of these patients was similar across the different KMT2A translocations, including those generating KMT2A/MLLT3 fusion; however, t(9;11)(p21.3;q23.3)/ KMT2A::MLLT3 AML had an almost significant improvement of RFS compared to other KMT2A translocations [ 25 ].
With respect to DFS, the later cohort showed a trend toward improvement, although the difference was not statistically significant (log-rank p = 0.176).
Consistently, further gating demonstrated a decreasing trend in PMN-MDSCs within the metastatic liver following paquinimod treatment ( Figure 4 E).
In the myeloid compartment, Ly6C/Ly6G profiling showed a trend toward reduced frequencies of Ly6G + cells in paquinimod-treated aged mice ( Figure 4 D).
In the present study, although the development of cAEs did not demonstrate a statistically significant impact on PFS, a numerical trend toward superior PFS was observed in patients who experienced cAEs compared with those who did not.
a negative trendgold
Assessment of cell counts and biochemical markers revealed a statistically significant negative association of baseline CRP (OR = 0.81, 95%CI: 0.63–0.98, p < 0.05) with CB6, as well as a negative trend for CAR (OR = 0.55, 95%CI: 0.24–1.02, p 0.06).
A two-sided p -value < 0.05 was considered statistically significant, and p < 0.01 was regarded as highly significant.
The association was statistically significant in the BRAF + MEK inhibitor (HR = 0.618, 95% CI: 0.429–0.891) and chemotherapeutic regimens (HR = 0.600, 95% CI: 0.405–0.889) subgroups, and marginally significant in the immunotherapy subgroup (HR = 0.672, 95% CI: 0.451–1.002).
These results establish daratumumab as the first agent to demonstrate an improvement in overall survival with a favorable trend in high-risk SMM, supporting a paradigm shift away from watchful waiting toward early intervention in this patient population.
Both markers demonstrated nominally significant differential expression and consistent survival associations and were therefore prioritized to construct the two-marker ICP2 signature.
sOPN showed a trend toward higher levels in advanced disease but did not reach statistical significance.
In China, the incidence of endometrial cancer is escalating, showing a significant trend towards diagnosis at younger ages [ 3 ].
