Notably, even after fitting the lead SNP in a conditional GWAS analysis, there were still other somewhat significant ( p < 1E–05) SNPs remaining for the HTFYslope trait (though not very strong signals; Fig. 2 ), suggesting that they could be other QTLs for heat tolerance, which were not captured by the lead SNPs identified in the study.
Principal findings Our analysis revealed that Mavacamten significantly reduced LVOT gradients across various clinical scenarios, with a pooled MD of −38.28 mmHg for post-exercise gradients and −59.02 mmHg for post-Valsalva gradients, both highly significant ( P < 0.00001).

The mean synonymous site nucleotide diversity ( π syn ) for X-linked genes (Tables 1 and 2 ; Additional file 1 : Table S4) is 0.00017, versus 0.0018 for all autosomal genes identified; the mean estimated nucleotide diversity for all sites ( π ) is 0.00038 for the X-linked region, again much lower than values for the autosomal region or PAR (0.0017 and 0.0020, respectively, which do not differ significantly (Wilcoxon rank sum P = 0.12), while the difference from the X-linked region is highly significant ( P < 1 × 10 −5 ) for both regions).
Relative to the codon frequency of the native DNA, the Mann-Whitney-Wilcox rank-sum analysis of the optimized CDSs shows that two are non-significantly (n.s.) optimized, four exhibit a p value between 1.4E−03 and 7.7E−04, and three are highly significant with a p < 1E−05 (GenScript, GeneArt, COOL).
