18 This well-powered meta-analysis produced one genetic variant of genome-wide significance (cholinergic receptor, nicotinic, alpha 5, CHRNA5 SNP rs2036527, P <5 × 10 −8 ), several additional variants that were approaching significance ( P <5 × 10 −7 ) and many variants that fell short of statistical significance for the prediction of cigarettes smoked per day.

The most statistically significant SNPs associated with late toxicity are listed in Table 5 , Table 6 , Table 7 , Table 8 , which show several SNPs had meta-p-values reaching or approaching significance (p-value ≤ 5 × 10 − 6 ) with concordant effect direction across the individual studies.

Additional SNPs approaching significance (p<1×10 −5 ) cluster within a 70 kb region of chromosome 10q23.31 that spans RPL7P34 and a gene called renalase ( RNLS ; MIM: 609360) ( figure 1B ).

Additionally markers on chromosomes 5, 10, 14, 15, 17, 21 and 24 had P values approaching significance (p≤5E-4).

Using this filtration strategy that leveraged the epigenomic data derived from the CMDGA, the Variant Sifter tool returned 79 variants, including one variant (chr9:504,491:A>C) demonstrating an association with ALS based on the meta-analysis of NDKP data that is approaching significance (β = 0.238, p = 7.01e-4; Figure 6D ).
