We then validated our findings using western-blotting for AATC, which showed a trend for decreased levels in SNCA 3x already at a basal level ( p = 0.2775), with a higher tendency noted upon the addition of TNF ( p = 0.1870) (Fig. 5H ), in line with the proteomics finding.
In the case of IL-5 and IL-10, 15% (11/73) and 16.4% (12/73) of patients diagnosed <10 years ago responded, respectively, while only 4.3% (1/23) of those diagnosed >10 years ago responded, which corresponded to a nonsignificant trend ( p = 0.283 and 0.179) in the same direction as observed for IFNγ.
Additionally, tracer uptake in the blood pool of symptomatic EAE mice was significantly higher than in naïve mice (p < 0.0001), with a trend toward significance in pre-EAE mice (p = 0.29; Supplementary Fig. 3a ).
Instead, we observed a weak trend towards prolonged survival in the KRAS-driven intestinal cancer model (median survival of 502 days ( VKras G12D/+ ;Snail KI/+ ) vs 354 days ( VKras G12D/+ ); p = 0.29; Fig. 2h, i ).
The percentage of potential T-cell responses achieved in each category revealed a nonsignificant trend toward higher responses among ACPA + RA patients to the created peptides (33%) than to no change (17%; P = 0.2918) and destroyed (13%; P = 0.1692) peptide groups, in addition to significantly more frequent responses among ACPA + RA patients to the created peptides (33%) than observed for the controls by χ 2 test (2.5%; P = 0.0002; Fig. 6b ).
Although the increase of oral Veillonella showed a trend toward higher esophageal colonization, this was not statistically significant (Fisher’s exact test, p = 0.303, n = 10; Supplementary Fig. 4L ).
