At the CD4SP stage, we found barely significant NCE enhancer function in the murine EL-4 CD4SP lymphoma cell line with the NCE1F, but not with the NCE1Fx2, even though the proximal enhancer was still functional ( Figure 2C ), indicating that NCE’s canonical enhancer function may diminish in mature CD4SP T cells.

MR analysis confirmed that downregulation of PRR4 expression is a risk factor for CRSwNP (OR = 0.99951, P = 0.04280), with the caveat that the very small effect size and borderline significance indicate limited evidence strength at present.
MR analysis preliminarily suggested that IL4R and IMPA2 may be potential risk factors for CRSwNP, while PRR4 may serve as a potential protective factor, though the effect sizes were modest (OR values close to 1) and P values were borderline significant (0.043–0.045), indicating limited evidence strength.
Although between-regimen differences were not statistically significant, mepolizumab 300 mg showed a numerical trend toward earlier glucocorticoid sparing (e.g., higher GC-free rates and fewer patients on higher GC doses at intermediate visits), which may reflect its preferential use in patients with systemic disease and its intended role in systemic disease management ( 6 , 7 ).

IgG antibodies reacting with AoC protein extracts were detected at both 14 and 35 days after AoC treatment alone or in the presence of OVA/alum, showing an increasing trend over time ( Figures 6B, C ).
In resident DCs, a decreasing trend was also observed at both 1–2 days post-injection, with no significant difference compared to PBS; a significant decrease was detected at 4 days post-injection ( Supplementary Figures 5A, B ).

Modern approaches illustrate an important trend toward integrated design and enhancement strategies that simultaneously optimize affinity, stability, and immunogenicity risk preserving developability attributes.
