A nominally significant interaction effect between the presence/absence of an alteration and efficacy of abemaciclib plus NSAI versus placebo plus NSAI was observed for EGFR , FGFR1 , CCND1 , and PIK3CA ( Fig. 3 ); however, these results should be interpreted with caution because of the exploratory nature of the analysis.
The nominally significant associations we identified showed no indication that non-HDL lipoproteins and related metabolic phenotypes increase EAA.
Although drinking status and diabetes showed nominally significant interactions, none of the interaction terms remained statistically significant after false discovery rate correction, suggesting that subgroup findings should be interpreted as exploratory rather than definitive.
This may explain, in part, why so many nominally significant medical research results are not borne out by subsequent studies.
In contrast, the CSF analysis revealed that disease progression was linked to increased levels of NAD+, arginine, saturated long-chain free fatty acids, diacylglycerides, triacylglycerides, and sphingomyelins, although these changes were only nominally significant [ 63 ].
Two of the eight most frequent genes in the top 14 pathways were also among the nominally significant genes in the gene-based FORGE analysis, i.e.
A single nominally significant coding SNP R311C (rs7314976, MAF = 0.14) within the GPR109A gene was taken forward for replication and found to be consistently associated with adiponectin in the three cohorts (CoLaus, Fenland and MRC Ely study, n = 8285, p = 4.6×10 −8 ) and HDL-cholesterol (HDL-C) in four cohorts (CoLaus, Fenland, Ely study and Lolipop, n = 18425, p = 2.9×10 −8 ) ( Figure S2A, S2B ).
We report the fraction of Bonferroni replicated pQTMs at 95% replication power and the fraction of nominally significant pQTMs with consistent directionality within all nominally significant pQTMs for all steps (Supplementary Data 3 ).
This increase was nominally significant at 56 dpi, but this should be viewed in the context of the broader trend towards increased expression.
A total of 54 lipid species (excluding TG and TG(O) species monitored by SIMs) showed a nominally significant interaction with sex in predicting change in WC ( Table S8 ).
For the brain caQTL, fetal brain mQTL, pancreatic islets sQTL and liver pQTL, only genome-wide significant or nominally significant results were available.
Mortality benefit was nominally significant with dapagliflozin but not with empagliflozin or sotagliflozin, raising questions regarding differences in baseline risk profiles or play of chance.
Cross‐product interactions We identified only one nominally significant interaction between MileAge delta and APOE genotype, suggesting that a one‐year higher MileAge delta was associated with 17% higher hazards of dementia in other diseases in the high vs low APOE risk groups (Table S22 ).
A P -value ≤ 0.05 before correction was considered nominally significant, but only corrected P -values below 0.003 are considered statistically significant in the interpretation.
Uncorrected p-values < 0.05 (nominally significant findings) were reported as showing a trend toward significance.
We mapped cis-QTL with QTLtools 1.3 39 to obtain (1) the top cis-QTL association for each gene after empirically adjusting p values using 1000 permutations, (2) all nominally significant associations, and (3) conditional cis-QTL associations after stepwise regressing out stronger, masking cis-QTL signals from the gene expression measurements.
When nominally significant associations were observed analyses were corrected for the False Discovery Rate (FDR) using Q-Value (bootstrap method) ( Storey, 2002 ). 3 Results No significant differences were observed between either COMT Met or COMT Val with respect to demographic variables with the exception of depression ( Table 1 ).
Of these 15, 11 were replicated in an external sample using strict criteria and the remaining four were nominally significant.
For CDKN2A/B rs10811661, we noted a nominally significant interaction between genotype and intervention; thus, we stratified diabetes incidence analyses by treatment arm.
Additionally, 10 % (6/58) of the nominally significant RD pairs showed heterogeneity, and 3 contained outlier SNPs (Additional files 3 and 4 ).
Both glycated products were more strongly correlated with the fasting (prebreakfast) and bedtime values than with the other time points, all but one of which were also nominally significant.
While none of the amines survived FDR correction ( q < 0.05), nominally significant findings are reported for hypothesis generation and should be interpreted cautiously pending validation in larger cohorts. 3.5.
This array was chosen because it contained pre-validated primer sets for three reference genes (Rplp1, Rpl13a, and Ldha) that were unchanged in our array data, six genes that showed a Benjamini-Hochberg corrected significant effect of treatment (Actb, Hspb1, Gfra1, Fgf9, Lifr, and Stat3), as well as several other neurotrophin-related genes that showed a nominally significant effect of treatment (e.g., Gfra2, Ngfrap1, and Npy) that did not survive FDR correction (see Methods section).
Three of the four methods yielded non-significant RD estimates consistent with the primary analysis, while the 1:1 PSM yielded a nominally significant result that likely reflects post-matching sample attrition.
