Using the parental transmitted and nontransmitted alleles in individual-level parent-offspring data from Iceland and Norway (deCODE, MoBa and HUNT; n = 46,105 parent-offsprings; Supplementary Table 11 ), we observed a nominally significant association between the maternal nontransmitted alleles polygenic scores for CBAT and testosterone and gestational duration.
Another gene that we found to be downregulated in the mouse GE is Eomes , which we also found to be specifically expressed in the human VZ and SVZ early in development (however note that expression of EOMES 8–9 pcw compared to all later stages was only nominally significant and did not survive correction for multiple testing) (Figures 3 , 4 ).
Although no homologous group was significant at False Discovery Rate (FDR)<0.05 (which appears to be too strict for comparisons across multiple species), by the more relaxed criterion of having a binomial p-value<0.05, four homologous groups were nominally significant: CAB39 , the genes encoding 14-3-3 proteins, sirtuins, and AHCY encoding S-adenosylhomocysteinase.
Studies in both postmortem and living human brain tissue have found limited correlations between methylation patterns in the brain and peripheral tissue of patients, with the highest interindividual correlation between nominally significant CpG sites in brain and blood at roughly 20%. 71 , 72 , 73 However, some analyses have shown that the correlation between methylation across brain and blood is stronger at certain CpG sites, and analyses of DNA methylation in blood could focus on these sites. 72 Studies of methylation in brain tissue from AUD cases rely on postmortem cohorts, which usually have a limited number of subjects.
