Subsequently, binomial tests were performed in R using the function ‘binom.test’, contrasting for each gene the number of nominally significant CpG-sites to the total number of probes annotated to each gene transcript not taking the direction of the methylation change into account.
Interestingly, the 'one-carbon pool by folate' pathway, which contains the MTHFR and MTHFD1L genes, was nominally significant for bipolar disorder, showed a trend for major depressive disorder and was not significant for schizophrenia, suggesting that this pathway has distinct effect on mood disorders, probably through a common intermediate phenotype, such as ruminative response style.
The Kolmogorov–Smirnov (KS) test (Table 1 ) revealed a nominally significant discrepancy in the distributions of exome-positive versus exome-negative cases of South Asian and Native American ancestries (reduced diagnostic yield associated with South Asian genetic ancestry, increased diagnostic yield associated with Native American genetic ancestry), however neither of these was significant after adjustment for multiple testing.
We defined a SNP as having a pleiotropic effect on two phenotypes when the P value for the O’Brien test statistic from the joint model of association of two phenotypes ( P joint ) with the SNP is at least one order of magnitude more significant than the P values ( P univariate ) for both phenotypes and the univariate P values are at least nominally significant ( P univariate < 0.05).
Furthermore, we specifically examined the 91 signature genes significantly associated with iPSCORE iPSC cardiac-fate outcome and found eight with nominally significant expression differences in the same direction (e.g., overexpressed or downregulated) in the two sets of iPSCs ( Figure 5 G), which is 2.5 times more than random expectation (p = 0.012, Fisher's exact test; Figure 5 H).
EX enatide S tudy of C ardiovascular E vent L owering Trial (EXSCEL) showed nominally significant reduction in ACM (but not for CV death) [ 3 ]; for ACM these studies appear to suggest that exenatide may be associated with mortality postponement in a broad population and certainly does not appear to increase morality risk.
Of the 32 variants included in the genetic score, 4 ( FTO , GNPDA2 , NEGR1 , and SEC16B ) showed nominally significant interactions with total fried-food consumption on BMI ( P for interaction <0.05) and all interactions were in the same direction (that is, stronger association between fried-food consumption and BMI in carriers of one or more BMI increasing alleles).
Accelerated biological ageing is not associated with cross-section and longitudinal measures of cognition In the cognitively unimpaired Aβ + cohort, a nominally significant association between cross-sectional PACC scores and accelerated ageing was observed with the Phenoage clock (Supplementary Table 1 , Additional File 1 ).
