Results Among pediatric subjects with WGS data, we identified a significant intronic variant in EDIL3 (rs149378, P = 1.9 × 10 −8 ) and a nearly significant intronic variant in LHX4 (rs79008224, P = 7.2 × 10 −8 ).
While this failed to surpass Bonferroni correction, the p -value across stages was highly significant (rs823128, p =7.3×10 −8 ) and it is worth noting that the significance improved for all SNPs at this locus when combining stage I and II results ( Figure 2C ).
3c ) and a less strong yet still highly significant directional effect of intelligence on BV ( b xy = 0.139, SE = 0.026, P = 7.34 × 10 −8 ; Fig. 3d ), suggesting a bidirectional association between these phenotypes, in line with the previous reports 53 .
Highly significant enrichment was uniquely found within the first quartile of ranked genes, including metabolic ( P < 7.4 × 10 -8 ), post-translational modification ( P < 9.7 × 10 -5 ) and developmental ( P < 1.3 × 10 -6 ) processes (all Benjamini-Hochberg adjusted).
Although no other SNPs reached genome-wide significance in the conditional analysis, a few borderline significant signals emerged: rs191288953 ( p =7.41E-08; chromosome 7; closest gene SDK1 ) and rs79935880 ( p =7.69E-08; chromosome 10; near MTRNR2L7 ) for C22:0 ceramide levels and rs34432959 ( p =8.68E-08) and rs12963655 ( p =8.00E-08) for C24:0 ceramide levels, both of which are located near SYT4 on chromosome 18.
In the secondary, combined meta-analysis, a total of 57 SNPs were associated with BDR at p<5×10 −6 ( supplementary table S2 ), 21 SNPs after linkage disequilibrium clumping ( table 2 ) and one SNP (rs4782614) associated with BDR as defined by per cent change in FEV 1 nearing significance (p=7.5×10 −8 ).
A third highly significant SNP ( p < 7.60 × 10 −8 ) was located on chromosome B04 (AX-147248780) at positions around 130738535 with 20 candidate genes within proximity, including several disease resistance proteins (TIR-NBS-LRR) ( Arahy.RGV79Z , Arahy.27BWBV , Arahy.QZS0X1 , Arahy.IB6E9K , Arahy.IB6E9K , Arahy.27QVSF , Arahy.EG26MQ , Arahy.
The clusters are highly significant in predicting survival (likelihood ratio = 37.0; p = 7.6×10 −8 ), also accounting for clinical and histopathological information (age, stage and cancer type; Methods), showing that considering the mutation data and their interactions provides a more complete and informative picture of tumours and their progression.
We observed a highly significant association of SNP rs7301743 in TBX3|MED13L with a 0.071 mV (95% CI, 0.038–0.11 mV) smaller voltage of the R wave in V5 per minor allele copy (P = 7.635 X 10 −8 ).
Meta-analysis of two independent Japanese datasets demonstrated a marginally significant association of SNP rs7301743 in TBX3|MED13L with a 0.071 mV (95% CI, 0.038–0.11 mV) shorter R wave amplitude in the V5 lead per minor allele copy (P = 7.635 x 10 −8 ).
Mega‐analysis of the effects of each APOE genotype compared with the ε3ε3 genotype showed highly significant associations for the ε3ε4 ( β = −1.41, p = 7.68 × 10 −8 ) and ε4ε4 ( β = −2.41, p = 3.32 × 10 −3 ) genotypes and suggestive‐effect significance for the ε2ε3 genotype ( β = 0.55, p = 8.26 × 10 −2 ) (Table 1 , Model 1). 2.3 Independent associations of TOMM40 and APOE polymorphisms with BMI Despite moderate LD between rs2075650 and rs429358 (Figure 1 ), bivariate mega‐analysis with these SNPs identified their independent associations with BMI, although their effects became smaller (Table 1 , Model 2).
