Although in this study a lack of association due to the low frequency of the variants was observed for 1007finsC and G908R polymorphisms 32 , a strong trend ( p = 0.05) towards a reduced presence of the mutated R702W allele was observed in IA patients 32 .

All signals show at least nominally significant ( P <0.05) directionally concordant associations in the follow-up studies of clinically validated PCOS cases, with no significant heterogeneity by PCOS case definition ( Supplementary Table 2 ).
Of these, 22,296 were nominally significant ( p < 0.05) and 4192 were significant to a genome-wide Bonferroni level ( p < 1 × 10 −7 , linear mixed model) for GWAS risk haplotype-capturing SNP allelic count.
We performed immunoprecipitation-mass spectrometry (IP-MS) identification of N-terminal-tagged CLK2-associated proteins (4 FLAG-tagged replicates, one untagged CLK2), using tandem mass tags (TMT) mass spectrometry and observed highly significant enrichment (Fig. 6a , Supplementary Data 32 ) of both known CLK-interacting proteins (28/97 known, B-H adjusted Wald p < 0.05) including CLK1 and additional splicing factors (98/270 total enriched, B-H adjusted Wald p < 0.05).
The minimum sample size requirement is based on experience and the fact that the observed differences between different conditions were statistically highly significant ( p <<0.05, Figs. 1 e, 2 d, 4 c, 7b ) and never at the borderline value of p = 0.05.
After mapping these CpG sites to genes, performing GO enrichment analyses, and removing overlapping GO terms, there were a total 95, 76, and 47 nominally significant (hypergeometric test P < 0.05) biological processes, respectively.
