Three further SNPs demonstrated nominally significant associations with preeclampsia: the intergenic SNP rs17249754 located just downstream of the ATPase, Ca 2+ transporting, plasma membrane 1 ( ATP2B1 ) gene ( P = 0.02, OR 0.80, CI 95% 0.66–0.97), the intronic SNP rs16933812 in the paired box 5 ( PAX5 ) gene ( P = 0.04, OR 0.85, CI 95% 0.74–0.99), and the intronic SNP rs12946454, located in the phospholipase C delta 3 gene ( PLCD3 ) ( P = 0.046, OR 0.85, CI 95% 0.73–0.80).
I could only find one cancer type (GBM) where mutations in TP53 are associated with a nominally significant increased age acceleration (p = 0.02)". as follows: TP53 mutation is associated with significantly lower age acceleration in six data sets (Additional file 6 ) including AML (P = 0.0041), breast cancer (P = 7.8×10 −12 and P = 1.4×10 −12 ), ovarian cancer (P = 0.04), uterine corpus endometrioid (P = 0.0012), and colorectal cancer (P = 0.036, Figure 4 B).

For multiple pregnancy rate ( Supplementary Table S4 ), while the pooled estimate was nominally significant (OR = 1.26, 95% CI: 1.03-1.55, P = 0.02), exclusion of three studies (Liang, Mao, and Wang) rendered the association non-significant, with 95% CIs crossing the null in all three scenarios; the effect, while directionally consistent, is statistically fragile and dependent on a subset of studies, with persistent high heterogeneity (I2 range: 61.42%-78.35%) further limiting confidence.
Given that several atherosclerosis risk factors have been associated with AS 6 , 18 , 19 , we tested the novel AS variants for association with the traditional cardiovascular risk factors and observed a nominally significant association ( P < 0.02) between rs1830321 and systolic and diastolic blood pressure in Iceland (Supplementary Data 3 ) and in data from the UK Biobank ( https://biobankengine.stanford.edu/search# ).
We further investigated the psoriasis/CAD genetic correlation by applying LDSC to our case-control psoriasis GWAS (11,024 cases and 16,336 controls) and a large CAD GWAS (60,801 cases and 123,504 controls), which provided a nominally significant ( p = 0.02) genetic correlation ( r g = 0.11).

Moreover, rs9313619 remained genome-wide significant when conditioning on rs29784 (conditional P = 3.50 × 10 −8 ), while if conditioning on rs9313619, the previously reported top SNP rs29784 remained only nominally significant ( P = 0.02).
There was a nominally significant main effect for enhanced VASC in the higher-dose group ( t (36) =−2.48, P =0.02), HCQ ( t (36) =−2.75, P =0.009) and STAI ( t (36) =−2.48, P =0.01), indicating more craving and state anxiety due to cortisol administration for the higher-dose group.
