Of these, only rs2736098 demonstrated a nominally significant pooled association but with a small effect size (OR: 1.25; 95% CI, 1.04–1.20) and high heterogeneity (I 2 = 87.1%) ( Fig. 6 ).

We identified 14 nominally significant correlations between BMD and our pleiotropic replicated phenotypes.
Specifically, the indirect effect of CAA and NFT on cognition is at least nominally significant when accounting for Lewy bodies, hippocampal sclerosis, vascular brain injury and arteriolosclerosis, with effect estimates consistently showing a reduction in cognition (faster decline).
Rigorous control of the type I error rate at the nominal level is of particular importance in SNP-trait association studies; given the multiplicity of hypotheses tested, nominally significant associations cannot be trusted unless type I error is strictly controlled.
Of note, DM–treatment interaction terms were not included given the insufficient number of events to detect subtle differences, with any resultant nominally significant P ‐values likely representing false‐positive results.
In addition, rare, potentially functional variants (MAF < 5%, Phred-scaled combined annotation dependent depletion score >10) showed a nominally significant enrichment in controls both: 1) across all five genes; and 2) in the F12 gene alone.
Our main effect analysis for migraine demonstrated only 6 nominally significant SNPs, and among them only one could be associated with a gene, the intron region of PRDM16 : rs2455107.
Gene set enrichment analyses (GSEA; Broad Institute 35 ) determined that nominally significant gene correlates of Cacna2d1 (Supplementary Data 1 ) were significantly enriched for plasma membrane localization (Fig. 2g ) and for functions related to neuronal function and excitability, including ion channel activity, substrate-specific channel activity, metal ion transmembrane transporter activity, ion transmembrane transporter activity, and receptor activity (Fig. 2h ).
One associated SNP from each significant or nominally significant locus and the clinical variables were included in a logistic regression model using IBM SPSS Statistics V20.
52 However, the association of HDL-C and TC was only nominally significant with BMI-dependent GRS2 in the current study.
However, we did find a number of nominally significant results, which are consistent with prior literature (see Supporting Information Table 3 , for full details of these results).
We also observed nominally significant sex-by-diagnosis interactions in the other four regions, but they did not survive FDR correction for multiple comparisons at the whole-brain level.
It has been reported that the prediction quality can be improved when there are a large number of SNPs, each of which is merely nominally significant, especially for diseases such as bipolar disorder and coronary heart disease[ 46 ].
Conclusion It is essential that formal procedures be adopted in order to avoid modestly positively results from candidate regions being swamped by the huge number of nominally significant results which will be obtained when very many markers are genotyped.
10 , 11 In AF, studies with a targeted approach have identified gene variants with nominally significant p values in the IL6R locus. 12 , 13 The use of genetic variants as instrumental variables (IVs) in Mendelian randomization (MR) eliminates reverse causation bias and allows causal inference.
Thirty-one 10-year metabolite changes showed a nominally significant T2D risk trend across quintile-based categories (P trend < 0.05) and were significant after multiple testing correction in the continuous analysis (FDR < 0.05, Wald test) ( Table 2 and Supplemental Tables 3 and 4 ).
Therefore, the significant results of CAPM-IADL (additive model) and nominally significant results of MIST-IMD (dominant model) in the same population are consistent with the minor A-allele of rs2253206 being correlated with better PM performance.
A further 428 proteins were identified to have evidence of colocalization [PP4 > 0.7] and at least a nominally significant MR association with the risk of cancer [min: 8 proteins for skin cancer and max: 241 for breast cancer] (Fig. 1 , Supplementary data 2 ).
The intraclass correlation coefficient for the paternal uncle-nephew pairs ( r = 0.65, n = 12, F = 4.64, P = 0.007, 95% confidence interval: 0.17, 0.88) and the combined paternal uncle-niece/nephew pairs ( r = 0.25, n = 43, F = 1.68, P = 0.047, 95% confidence interval: −0.05, 0.51) were nominally significant.
The effect of empagliflozin on first and recurrent hospitalizations for heart failure was consistent in 12 prespecified subgroups, except for a nominally significant treatment-by-race interaction ( Figure II in the Data Supplement ).
84 were nominally significant, but after FDR correction, 27 remained significant.
A study analyzing gene-based associations between the GRN , TMEM106B , ABCC9 , and KCNMB2 genes and HS found Bonferroni-corrected significant associations for ABCC9 assuming a recessive mode of inheritance (MOI) and nominally significant associations with GRN , TMEM106B , and KCNMB2 [ 29 ].
Identified metabolites showing a sex‐specific response to 17aE2 We further assessed changes in individual known metabolites with 17aE2 treatment, again focusing on metabolites that change specifically in males but not females, as indicated by a significant two‐way interaction between sex and treatment, and also showing a nominally significant response to 17aE2 in males.
Prespecified analyses in PROOF-HD showed clinically meaningful and nominally significant benefits and improvements from baseline in disease progression, as well as motor and cognitive outcome measures when comparing pridopidine with placebo.
P -values of < 0.05 were considered nominally significant.
