Marker-by-marker analyses of rs1800888, rs1042718 and rs1042719 of ADRB2 in the present study and the previous published analyses of rs1042713 and rs1042719 [4] showed nominally significant association with waist-to-hip ratio and systolic blood pressure in line with the results seen for haplotype pairs.
The associations for the Hannum clock were not statistically significant from any of the meta-analyses, and the rest of the associations were found to be nominally significant in at least one of the meta-analyses, with the exception that the association between number of live births and genome-wide average DNA methylation was marginally significant ( P = 0.07).
In support to the pathophysiological roles of the stress score genes, pathway enrichment (gene ontology) analysis of all nominally significant differentially expressed genes between T1D macro- and normoalbuminuria groups also suggested changes in oxidation-reduction as well as transporter and EV-linked activities (p < 0.05, logFC>1, n = 130 genes, Table S8 ).
There was no association with fracture overall (odds ratio [OR] = 1.01 [CI, 0.95 to 1.08]) in either unadjusted or adjusted analyses, but there was a non-significant trend for association with vertebral fracture and a nominally significant association with incident vertebral fractures in females (OR = 1.33 [CI, 1.00 to 1.77]) that was independent of BMD, and unaltered in adjusted analyses.
As this work is exploratory in nature, the number of comparisons conducted relative to the available sample size limits the interpretability of individual P values, and many nominally significant associations do not survive multiple testing correction; nonetheless, the consistency and strength of the associations observed with cardiac fibrosis are notable and warrant further investigation in larger cohorts.
