A nominally significant association with cancer overall was observed for homozygosity at HLA-DQB1 locus (adjusted OR = 1.08, 95% CI = 1.01–1.15, P = 0.016, Table 2 ).
We also observed nominally significant associations of several genes with CAD: LPL [ 29 , 60 ] (OR = 1.168; CI 1.034–1.036; P = 0.016), NOS3 [ 15 ] (OR = 1.143; 95% CI 1.109–1.279; P = 0.02), and ADAMTS7 [ 32 ] (OR = 1.062; 95% CI 1.011–1.115; P = 0.016) (Supplementary Tables 9–10).
In Period 1–6, the SIDD subgroup had 12% (2; 22) lower HOMA2-B value (reflecting insulin secretion at fasting) than controls ( ESM Figure S6B, S21 ), but the difference was only nominally significant (p = 0.016).
Association tests adjusted for age and sex revealed a nominally significant association using logistic regression between AMD and three SNPs ( DAPL1 : rs17810398:C>T, P = 0.016; RP1 : rs9643828:T>C, P = 0.037; CST3 : rs2424577:C>T, P = 0.028) (Supplementary Table S2).

3.3 Identification of differentially expressed miRNAs Of the sixteen reliably detected miRNAs, three, miR-15b, miR-132, and miR-652, showed a nominally significant increase in expression in the high-risk subjects compared to the control subjects (miR-15b: p = 0.0166, fold-change (FC) = 1.29; miR-132: p = 0.0249, FC = 1.34; miR-652: p = 0.0108, FC = 1.26; Fig. 1 ; Table 2 ).
