The FDR-corrected statistical analysis (Benjamini–Hochberg procedure) reduced the number of nominally significant findings from 37 to 34, with three borderline parameters (cobalt, mercury, beryllium) reclassified as nominally significant pending replication.
In internal replication, all the newly identified loci were nominally significant both in East Asian and European populations (Supplementary Fig. 7b and Supplementary Data 25 ). rs35593046 ( CACNA1D ), rs7659823 ( EDNRA ), rs5823966 ( ZNF397 ), and rs1541596 ( CARM1 ) were much more frequently observed in the East Asian population (Supplementary Fig. 7c and Supplementary Data 25 ).
GLM analyses revealed nominally significant differences between the aADHD and HC groups in several domains, however, only the Rapid Visual Information Processing measures survived correction, indicating impaired sustained attention and response inhibition in the aADHD group.
86 , 87 In addition, although pegcetacoplan-treated patients with GA secondary to AMD in the phase III DERBY trial ( NCT03525600 ) narrowly missed the primary endpoint of reduction in GA lesions at Month 12 of the study, 86 longer-term analysis of patients has demonstrated that pegcetacoplan contributed to nominally significant reductions in GA lesion growth at Month 18. 87 Pegcetacoplan was also associated with a 73.3% reduction in proteinuria in five patients with C3 glomerulopathy in the phase II DISCOVERY trial ( NCT03453619 ) 88 and demonstrated preliminary efficacy in patients with cold agglutinin disease in the PLAUDIT trial ( NCT03226678 ). 89 It is likely that pegcetacoplan’s approved usage will be expanded to some of these conditions in the future, although the dosage and route of administration remains to be determined.
Dyslipidemia (measured through cholesterol, triglycerides, HDL, LDL) A prior study assessing CAD risk scores for 2,599 participants of the Dutch-Belgian Lung Cancer Screening Trial revealed nominally significant associations for genetic risk scores of low-density lipoprotein-cholesterol, total cholesterol, and obesity [ 48 ].
We regarded a locus as being fully replicated if the locus from the original GWAS study (defined as the genomic risk locus ± 100 kb) overlapped with a genomic risk locus in any of the latent factors ( P GWAS < 5 × 10 −8 ) or partially replicated if it demonstrated a nominally significant association with one of the latent factors ( P Bonferroni < 0.001 (that is, 0.05/47)).
