An RNA LASSO model consisting of 25 genes was found to predict survival in PAH; 372 of the 507 top dysregulated genes from the whole blood RNAseq study were present in the lung tissue microarray study 44 and 161/372 (43%) were also directionally consistent; 41/161 (25%) genes were nominally significant and 26 met false discovery rate (FDR) corrected significance.
When suicidal behaviour was defined as “suicide, MedDRA preferred term for suicide attempt or suicidal ideation, or the standardised MedDRA query for suicide/self-injury,” the unadjusted association is nominally significant, and the direction of the association is the same (OR: 0.57; 95% CI 0.33–0.97 per number of minor alleles) ( Table 3 ).
Features, nominally significant in either of these two models, were further filtered out if not matching the suggested metabolite biomarker candidates, first by MS (if, e.g., matching to an isotope instead of the main fragment) and later by MSMS by comparing fragmentation patterns to the Human metabolome database hits of the suggested biomarker candidates.
We observed two nominally significant Reactome pathways with the genes with fold change ≤ 0.74 and P adj < 0.001: Glucuronidation and Biological Oxidations ( P adj = 0.04 for both pathways), and 17 enriched Reactome pathways with the genes with fold change ≥ 1.34 and P adj < 0.001 ( Figure 3B ).
Across all three eosinophil risk categories, no statistically significant between-group differences were observed at either timepoint, with the exception of a nominally significant finding in the high-risk category (≥300 cells/μL) at both 6 and 12 months; as these counts fell below the TriNetX privacy threshold and were reported as rounded values, no conclusions regarding eosinophilia trajectory can be drawn from these data ( 32 ).
