Robust gene signatures of kidney aging and their correlation with functional and structural dimensions of kidney health Of 19 replicated genes, 14 showed at least 1 nominally significant association with 1 of 5 biochemical or histologic measures of renal functional and structural integrity known to deteriorate with age (i.e., estimated glomerular filtration rate, glomerulosclerosis, tubular atrophy, interstitial fibrosis, and arterial/arteriolar narrowing) in up to 160 individuals from the TRANSLATE Study ( Supplementary Tables S10–S14 , Figure 2 , and Supplementary Figure S2 ).
Although we report both nominally significant and biologically plausible trends to support hypothesis generation, many of these did not reach Bonferroni-corrected significance.
The following results therefore describe nominally significant patterns and directional trends, which should be interpreted as hypothesis‐generating rather than confirmatory findings. 3.4.
A p < 0.05 was reported as nominally significant, and a Hochberg correction ( Hochberg, 1988 ; Huang and Hsu, 2007 ) was applied to determine statistical significance in the context of multiple comparisons within a given genotype and time-period of interest.
In the final model we retained all nominally significant predictors.
An association test was performed for each SNP and the number of nominally significant tests in a segment was counted.
In the phase 2 study of tirasemtiv, no efficacy signal was seen in ALSFRS-R, although a key secondary end point, SVC, showed a nominally significant benefit. 9 , 11 , 12 SVC was the primary end point of the phase 3 tirasemtiv study; negative results were believed to reflect poor tolerability, as a per protocol post hoc analysis again suggested a benefit. 11 In the FORTITUDE-ALS trial, the phase 2 study of reldesemtiv, SVC was the primary outcom
For the secondary endpoint of time to viral clearance, despite randomization being a median of 11 days after symptom onset for both opaganib and placebo cohorts, opaganib demonstrated a nominally significant improvement as compared to placebo, with a hazard ratio of 1.34 and a nominal p -value of 0.043 as shown in Figure 2 (and Table 3 ).
The FDR-adjusted P was presented by “ q ”, and the results were considered as nominally significant if q < 0.10.
23 The effect of fully CSF confirmed versus mixed or non-CSF confirmed cohorts influenced the prevalence of behavioural symptoms only, however, the association was nominally significant ( Supplementary Table 15 ).
Changes in brain vMRI (ml, %) detected by boundary shift integral (BSI) 42 , an analytic method employed in other HD studies 21 – 23 , 43 , showed a nominally significant 26% reduction in caudate atrophy for EM subjects (PEPI, n = 90; PBO, n = 88) treated with pepinemab relative to placebo, and the percentage change from baseline at month 17 resulted in LS mean difference (95% CI) = −1.54 (−2.79, −0.29) (two-sided P = 0.017; Fig. 2a and Extended Data Table 4 ).
A recent study demonstrated that combining genes with nominally significant GWAS P values and tissue-specific networks were powerful in building machine learning classifiers for identifying novel genes associated with disease [ 57 ].
We identified 11 nominally significant associations linking cell types to SS.
Throughout the results section, nominally significant results are reported.
In our analyses, CD55 was also identified as a significant DEG in dendritic cells and monocytes, and showed nominally significant associations in eQTL-based MR.
Overall, 35 (62.5%) of the associations were nominally significant.
Statistical power for the nominally significant MR estimates was calculated using the mRnd online calculator ( https://shiny.cnsgenomics.com/mRnd/ ), incorporating the sample size of the outcome GWAS, the proportion of variance in the exposure explained by the IVs (R2), and the observed odds ratio, with a Type-I error rate (α) set at 0.05 [ 33 ].
Our results, although nominally significant, were not significant after correcting for multiple comparisons.
Ninety-eight of the six hundred nine investigated microRNAs showed nominally significant P -values, suggesting that BD-associated microRNAs might be enriched within known microRNA loci.
All statistical tests were two-sided, with p -values < 0.05 considered nominally significant.
Although only one estimate was still significant after the Bonferroni correction, other nominally significant estimates should also be treated carefully.
In fact, the study of nominally significant genomic loci in the meta-analysis has shown that pathways related to aggregated amyloid-β, tau protein, and inflammatory pathways could be related to PH occurrence. 2.
A cut-off p value <0.01 was used to retrieve nominally significant SNPs.
The cytochrome P450, cell cycle control, and alcohol dehydrogenase candidate genes groups also exhibited nominally significant (0.01<P permuted <0.05) associations with smoking behaviors ( Table 4 ).
