Overall selectivity of KT777 knockdown ( KCNT1 downregulated at an adjusted P value ( P adj ) = 1.7 × 10 −34 ; 11 additional downregulated genes nominally significant at P adj < 0.05; Supplementary Table 5 ) was similar to that of RNase H-activating ASOs BIIB067 and HTTRx (targeting SOD1 and HTT , respectively) (Supplementary Figs. 1 and 2 ).
SKAT required 15FC × 10FNC to report nominally significant p -values, GSKAT required 20FC × 5FNC to report statistically significant p -values, FarVAT-CALPHA did not generate significant p -values unless we used the BLUP correction; FarVAT SKATO reported p -values that were significant at 15FC × 10FNC, and at 5FC × 20FNC if we used the BLUP correction.
In a study using the TEDS sample at age 7 ( Meaburn, Harlaar, Craig, Schalkwyk, & Plomin, 2008 ), an initial discovery stage based on “pools” of DNA from low- and high-reading-ability groups ( n = 750 in each group) identified 10 nominally significant SNPs that replicated in a second TEDS subsample of individually genotyped individuals.
To further assess the relevance of driver meQTLs to cancer risk, we used them to predict breast cancer status alongside clinical covariates using the approach described by Elgart et al . 18 We first performed feature selection by LASSO on nominally significant driver meQTLs and available clinical factors (age, ancestry as represented by the top 10 genotype-derived PCs); LASSO regularization removed ancestry and some meQTLs.
Discussion Genes harbouring markers with only modest evidence of association (nominally significant but not reaching the genome-wide significance threshold) can be identified if they belong to the same biological pathway or mechanism; therefore, pathway-based approaches, which jointly consider multiple variants in interacting or related genes, might complement the most-significant SNPs/genes approach for interpreting genome-wide association (GWA) data on complex diseases [ 7 , 8 ].
In the RCT, nominally significant placebo-adjusted differences in LS mean change from baseline to end point on the ADHD-RS-IV were observed in GXR-treated participants, in both subgroups (prior MPH, −9.8 [95% CI −14.6 to −5.1], P <0.001, ES 0.85; stimulant-naïve: −7.6 [95% CI −11.8 to −3.3], P <0.001, ES 0.65).
