GV improvements at 6–12 months were also uniform across most subgroups, although a nominally significant interaction was observed for AEs (P interaction=0.02), with patients without AEs showing slightly larger GV improvements (1.316 vs. 0.792 cm/year).
In GWAS meta-analysis of stage 1 cohorts, the minor T allele of rs12917707 at the UMOD locus, previously identified by GWAS to be associated with higher eGFR in cross-sectional analysis [ 14 ], was associated with an increase in eGFR over time at a genome-wide significant level (p=2.6×10 −14 , Table 2 ), and showed at least nominally significant, direction consistent association with all other analyzed phenotypes ( Supplementary Table 4 ).
We used 33, 8, 12, and 4 risk allele SNPs which were nominally significant in BMI, WC BMIadj , WHR BMIadj , and BF% GWAS results in JHS, to characterize individual variants’ association with BF%, SAT, VAT, and VSR ratio, respectively; each SNP represents an independent locus within the genome (based on linkage disequilibrium).
When we modeled FEV1 and FVC exclusively with height, the effect of height on CAD risk did not appear to be reduced by lung function (OR = 1.13, 95% CI = 1.10–1.16, p = 3.2 x 10 −17 , Fig 3E ), although the effects for both lung function measures were nominally significant at best (p FEV1 = 0.026 and p FVC = 0.052) Fourth, we created a restricted set of height genetic instruments that excluded variants nominally associated with FEV1 or FVC [ 24 ] (p < 0.05) to more directly compare between results from the published work suggesting lung function’s effect on height [ 5 ], and only retained variants present in lung function, height, and CAD GWAS.
In the overall study population, and in those considered to have cardiac involvement (based on the prespecified definition used in the post hoc APOLLO analysis), treatment with vutrisiran was associated with beneficial changes in NT-proBNP and a nominally significant beneficial change in stroke volume compared with placebo.
