Additionally, 22 SNPs including C8B rs2281598, C8B rs72670362, C8B rs2269113, C8B rs582317, C8B rs614020, C8B rs2236217, TRAIL/TNFSF10 rs231987, FKBP5 rs9357201, FKBP5 rs2766533, FKBP5 rs9348981, SFRP1 rs17574424, APOA4 rs5104, APOA4 rs5100, APOA4 rs5092, APOA1 rs7116797, LGMN rs4904981, LGMN rs1242108, LGMN rs1015033, LGMN rs68062833, LGMN rs1242102, MFG-E8 rs10859, MFG-E8 rs17202544, MFG-E8 rs555006940, MFG-E8 rs2271715, and ANGPTL4 rs2278236 showed nominally significant association with TN (p for all < 0.05), and none of them remained significant after Bonferroni correction (as shown in Table S1 ).
Interestingly, two of the candidate genes, namely PDE2A and PIK3C2B , were reported with nominally significant P -values for the association with NAFE in the Epi25 collaborative, the largest WES study in epilepsy to date. 38 While we did not find any additional URV in PDE2A , two heterozygous PIK3C2B URVs were identified in FE cases: a stop-gain p.Q1533* variant and a missense p.I1544 M variant in the C2 do
This effect is consistent with a nominally significant association between decreased NF1 subcutaneous adipose expression and decreased endometrial cancer risk in our recent TWAS. 22 NF1 encodes neurofibromin (NF1), a large multifunctional tumor-suppressor protein that is involved in several cell signaling pathways and regulates many cellular processes such as proliferation and migration. 28 NF1 is also associated with neurofibromatosis type 1 (MIM: 162200 ), a Mendelian disease characterized by fibromatous skin tumors.
Furthermore, polygenic risk score (PRS) analyses are based on the premise that hundreds, or even thousands, of loci contribute to the inheritance 1 , 46 , 47 , and cross-inheritance 48 of psychiatric diseases, but in PRS, it is unclear which of the nominally significant SNPs represents a contributing locus based on the statistical criteria by which they were selected.
In the correlation analyses, we found that the SAT bulk expression of the lncRNA, AC002066.1 , was significantly positively correlated with the adipocyte diameter in a BMI-dependent way as a significant correlation was observed in the lower BMI twin siblings from each BMI-discordant MZ pair (adipocyte diameter: Spearman’s rho = 0.55, p adj = 0.0023) ( Figure 5 E; Table S13 ), while the higher BMI twin siblings from each BMI-discordant MZ pair showed only nominally significant p values ( Figure 5 D; Table S13 ).
