For traits in which the exposure–outcome associations were nominally significant in all four estimators and reached test-wide significance ( P < 0.05/9) in two out of four (Supplementary Table 20 ), we applied the outlier-robust and pleiotropy-robust estimators MR-RAPS 94 and MR-PRESSO 95 .
However, the nominally significant correlation between the ΔMI map and the U-T axis map was not robust to significance testing using the spin-test method to control for spatial autocorrelation in the maps, and the correlation between the ΔMI map and the U-T axis map was evidently not as strong as the correlation between the MI map and the U-T axis map.
Although this GWAMA also included participants from other ethnic groups, the direction of association for the same reference alleles were largely similar between the South Asian and multi-ethnic samples (concordance of 70% and 92% for all variants, and nominally significant SNPs respectively, data not shown) 9 , which substantiates the use of this dataset.
Adjusted for imbalances in baseline characteristics or nonadherence, a nominally significant reduction in the primary composite end point was found. 41 Additionally, further prespecified secondary analyses of EVOLVE demonstrated a significant risk reduction for parathyroidectomy 40 or the development of calciphylaxis 42 with the use of cinacalcet.
Exome-wide burden of recessive genotypes In autosomes, there was no significant excess of recessive genotypes in probands after correcting for multiple testing (homozygous, compound heterozygous and all recessive genotypes, two variant types and two MAF thresholds), although there was a nominally significant excess in probands of rare (MAF ⩽1%) nonsynonymous compound heterozygosity ( Table 1 ).
