The effect of allocation to rt-PA on stroke outcome in patients ≤80 and >80 years of age will also be estimated (again, adjusted for other baseline covariates and treatment modifying effects of those covariates) and, if evidence of independent effect modification by age is observed, a test for difference in the two log-odds ratios will be performed (by comparing the difference in log odds ratios divided by its standard error against a standard normal distribution, a test that will be deemed nominally significant if the two-sided P -value is less than 0·05).
When the Bonferroni-corrected p-value threshold was less than 0.017 (correcting for 3 exposures and 1 outcome), the correlation was considered to have statistical significance, while a p-value less than 0.05 was regarded as nominally significant evidence of a potential causal association ( 46 ). 3 Results 3.1 Assessment of immune infiltration in OA patients Given the importance of changes in the immune microenvironment contributing to the progression of OA, we compared the immune scores of OA and normal individuals in the discovery dataset, which provided a qualitative assessment of the immune microenvironment ( Figure 2A ).
None (lead replication SNP: rs860447, MAF = 0.26, p = 0.60, Yale-Penn/STARRS EA-EA meta p = 6.4 × 10 −3 ) replicated in the STARRS EA data, but 34 of the top 98 EA SNPs were nominally significant in the STARRS LAT samples, and, remarkably, all 98 had effect direction consistent with what we observed in Yale-Penn EAs; the lead SNP from Yale-Penn EAs was nominally significant in STARRS LATs (rs1677091, MAF = 0.28 p = 6.52 × 10 −3 ) and improved in a meta-analysis of the Yale-Penn EA and STARRS LAT samples (meta p = 4.7 × 10 −9 ).
