It is interesting to note that the recently reported Genetic Association Information Network (GAIN) consortium study of recurrent depression reported nominally significant evidence for association at CACNA1C, 24 although it is important to recognize that the genotyping platform (and hence, single nucleotide polymorphism set) differed from that used in the WTCCC analysis.
For each transcript, we counted the number of experiments where the significance for differential expression was ≤0.05, and then generated the binomial probability, referred to as the consensome p-value (CPV), of observing that many or more nominally significant experiments out of the number of experiments in which the transcript was assayed, given a true probability of 0.05.
41 GWAS results for SRE remain inconclusive so far due to relatively small sample sizes but have included nominally significant associations between initial alcohol sensitivity and a genetic marker in GABRA6 . 44 Several studies have also identified associations between alcohol dependence and GABAergic SNPs on chromosome 4 ( GABRA2 , 33 , 45–50 GABRA4 , 16 GABRB1 , 51 GABRG1 42 , 49 ) and chromosome 5 ( GABRA1 , 43 , 52 GABRA6 , 53 , 54 GABRG2 33 , 55 ).
Associations of SNCA, GBA1, and UGCG Expressions with Depression Severity and Anxiety Based on the elevated gene expression levels observed in depressed patients compared to unaffected individuals, we conducted an exploratory analysis to determine whether high gene expression levels correlated with depression severity as assessed by clinicians (HAM-D, MADRS) or self-evaluation (BDI-II) with subsequent sex-stratified analysis in the event of nominally significant effects.
