Metabolites Associated With DR We found 17 metabolites associated with DR without adjustment for confounding factors; 7 of them were statistically significant ( P < 5.0 × 10 −5 ) and 10 nominally significant ( P < .006) after the adjustment for confounding factors.
Of the three loci identified in the main analysis, the locus on chromosome 11 was nominally significant in the Chinese cohort ( P = 0.006, Supplementary Fig. 8C ).

The SECA primary test for the overlap of associated effects, including all 144 SNP subsets, revealed more subsets than expected by chance showing at least nominally significant pleiotropy between endometriosis and endometrial cancer ( P = 6.0 × 10 −3 ): The pair of SNP subsets producing the minimum exact binomial test P ‐value for pleiotropy (endometriosis SNP subset with P ≤ 0.002 and endometrial cancer SNP subset with P ≤ 0.86) had P = 3.3 × 10 −4 .
Again using the thresholds ( p value <0.05 and Δβ ≥ 0.1) analysis revealed one nominally significant mQTL for FAM63B_1 (rs4774353 in FOXB1 , chr15:60309674 (hg19), with p value = 0.006 and Δβ = −0.11 for the A allele).
However, the study reported a nominally significant reduction in the rate of exacerbations (rate ratio 0.58, 95% CI 0.39–0.85; p=0.0061) and improved lung function among former smokers with itepekimab versus placebo [ 159 ].
Unlike singleton disruptive variants, although the burden of rare disruptive variants in SCZ was nominally significant, and survived FWER correction for all tests within the trait, it did not meet the threshold for tests across all traits (unadjusted P =0.0061, FWER within P =0.0228, FWER across P =0.0863, Supplementary Table S9 ).
In addition, damaging or rare damaging NOD2 variants were nominally significant across all three burden tests applied (p = 0.0063 to 0.045).
We also observed nominally significant results in relation to SCZ: a positive correlation between SCZ GWAS results and HB scores for complete selection (p = 6.37*10 −3 , S1 Table ; permutation p = 3*10 −3 , S2 Fig ).
Nonetheless, the lead SNP at the PAX1 locus showed a nominally significant interaction (β_int = 0.0309, P = 6.6 × 10 − 3), mirroring the direction and significance of the heterogeneity statistic.
The radial glia-like cells (RGCs) in the subventricular zone (SVZ-RGCs, p = 0.0035), RGCs in the dentate gyrus (DG-RGCs, p = 0.0261), and oligodendrocytes (p = 0.0306) showed nominally significant enrichment in the cell-specific enrichment analysis ( Figure 4 B), of which DG-RGCs were also nominally significant (p = 0.0066–0.0484) in three robustness analyses ( Table S10 ).
Specifically, tumors from chemosensitive patients showed nominally significant enrichment in natural killer (NK) cells ( p = 0.0069, BH adj. p = 0.1596), CD4+ Th1 T cells ( p = 0.0089, BH adj. p = 0.1596), and CD4+ non-regulatory T cells ( p = 0.0290, BH adj. p = 0.3423).
Of note, a signal near SLC26A9 , a known cystic fibrosis modifier gene 54 , was highly significantly associated with PEF in UK Biobank (P=3.97×10 −66 ) and nominally significant in SpiroMeta (P=6.93×10 −3 ), with consistent direction of effect, but did not meet the Tier 2 criteria.
