Another recent study analysed the association between SNPs from 46 previously reported adult height loci in children and found nominally significant associations for 15 loci ( 16 ), but that study analysed together all children between the ages of 0 and 18 years (preventing identification of age-specific effects) and does not have repeat measures on the same children.
Of these, 122 were autosomal SNPs, of which we selected 35 common SNPs (minor allele frequency >5%) that had at least a nominally significant and directionally consistent association with 25(OH)D in an earlier independent GWAS by the Study of Underlying Genetic Determinants of Vitamin D and Highly Related Traits (SUNLIGHT) Consortium, which did not include the UK Biobank ( 32 ) ( Supplemental Table 3 ).
For each immune cell type, nominally significant genes were partitioned into up-regulated and down-regulated sets and submitted separately to identify perturbagens whose transcriptional signatures were either concordant (“Up”) or inversely aligned (“Down”) with the observed expression patterns.
For new or worsening nephropathy there was a nominally significant quantitative interaction with a p-value of 0.0013 due to an inconsistent pattern of treatment effect (U-shaped pattern with lowest HRs of 0.45 and 0.42 in patients without MetS and those fulfilling 4–5 criteria compared to HRs of 0.81 and 0.71 in those fulfilling 2 or 3 criteria, respectively).
Consistently, the HRs from the adjusted model for the comparison of ET versus ribociclib (HR = 1.288; 95%‐CI: [1.006; 1.649]) or CT versus ribociclib (HR = 1.776; 95%‐CI: [1.377; 2.290]) were in favor of ribociclib, and the related 95%‐CIs indicated nominally significant differences.
