No correlation reached the significance threshold of p < 0.05 after adjusting for depressive symptom severity, age, sex, and years of education, although some nominally significant correlations were observed in a few brain regions before adjustment.
In a study using the TEDS sample at age 7 ( Meaburn, Harlaar, Craig, Schalkwyk, & Plomin, 2008 ), an initial discovery stage based on “pools” of DNA from low- and high-reading-ability groups ( n = 750 in each group) identified 10 nominally significant SNPs that replicated in a second TEDS subsample of individually genotyped individuals.
Markers associated with phenotype traits either at ‘close to genome-wide significant’ p-values or at ‘nominally significant’ p-values Results from tests of association pointed to only one risk variant (namely Chr15:40531386-rs12440118 from ZNF106 ) associated with HbA1c emerging at ‘close to genome-wide significant’ p-value of 3.41E-08 (with Benjamini–Hochberg FDR p-value of 0.0032) ( Table 2 ).
SKAT required 15FC × 10FNC to report nominally significant p -values, GSKAT required 20FC × 5FNC to report statistically significant p -values, FarVAT-CALPHA did not generate significant p -values unless we used the BLUP correction; FarVAT SKATO reported p -values that were significant at 15FC × 10FNC, and at 5FC × 20FNC if we used the BLUP correction.
Overall selectivity of KT777 knockdown ( KCNT1 downregulated at an adjusted P value ( P adj ) = 1.7 × 10 −34 ; 11 additional downregulated genes nominally significant at P adj < 0.05; Supplementary Table 5 ) was similar to that of RNase H-activating ASOs BIIB067 and HTTRx (targeting SOD1 and HTT , respectively) (Supplementary Figs. 1 and 2 ).
We identified a nominally significant interaction between total zinc intake and the SLC30A8 rs11558471 variant on fasting glucose levels (β-coefficient ± SE per A allele for 1 mg/day of greater total zinc intake: −0.0017 ± 0.0006 mmol/L, summary interaction P value = 0.005); this result suggests a stronger inverse association between total zinc intake and fasting glucose in individuals carrying the glucose-raising A allele compared with individuals who do not carry it.
Briefly, the predefined statistical criteria used to identify biomarkers of potential interest included: a fully adjusted, significant (false discovery rate of ≤0.05) prospective association with VTE in the tofacitinib treatment arm(s) or a nominally significant association in conjunction with a fully significant treatment effect (tofacitinib vs TNFi) on the change in biomarker levels in a direction matching the association with VTE risk.
