Robust gene signatures of kidney aging and their correlation with functional and structural dimensions of kidney health Of 19 replicated genes, 14 showed at least 1 nominally significant association with 1 of 5 biochemical or histologic measures of renal functional and structural integrity known to deteriorate with age (i.e., estimated glomerular filtration rate, glomerulosclerosis, tubular atrophy, interstitial fibrosis, and arterial/arteriolar narrowing) in up to 160 individuals from the TRANSLATE Study ( Supplementary Tables S10–S14 , Figure 2 , and Supplementary Figure S2 ).
EXSCEL reported a nominally significant reduction in all-cause mortality (HR: 0.86, 95% CI: 0.77-0.97) despite a neutral primary efficacy result [ 17 ], and PIONEER 6 showed lower cardiovascular and all-cause mortality (all-cause death, HR: 0.51, 95% CI: 0.31-0.84), albeit on a small number of events [ 19 ].
In the phase 2 study of tirasemtiv, no efficacy signal was seen in ALSFRS-R, although a key secondary end point, SVC, showed a nominally significant benefit. 9 , 11 , 12 SVC was the primary end point of the phase 3 tirasemtiv study; negative results were believed to reflect poor tolerability, as a per protocol post hoc analysis again suggested a benefit. 11 In the FORTITUDE-ALS trial, the phase 2 study of reldesemtiv, SVC was the primary outcom
APOB, APOC3 and its subfractions (heparin precipitate, heparin-soluble [HS]) and apolipoprotein-defined lipoprotein subclasses–classified Lp-B were associated with any-CVD in unadjusted analyses, and association of APOC3-HS remained nominally significant in adjusted analyses.
In the RCT, nominally significant placebo-adjusted differences in LS mean change from baseline to end point on the ADHD-RS-IV were observed in GXR-treated participants, in both subgroups (prior MPH, −9.8 [95% CI −14.6 to −5.1], P <0.001, ES 0.85; stimulant-naïve: −7.6 [95% CI −11.8 to −3.3], P <0.001, ES 0.65).
18 However, this inconsistency was nominally significant, and a complementary analysis confirmed benefits of semaglutide versus placebo across a continuum of BMI values. 18 As the current pooled post hoc analysis included data for OW s.c. and OD oral semaglutide from the SUSTAIN 6 and PIONEER 6 CVOTs, respectively, our findings on consistent risk reduction of CV outcomes regardless of baseline BMI support previous conclusions.
