The samples have been split into two sets in order to save DNA and reduce genotyping costs: the first set ( n =2271 cases and 2280 controls) is genotyped for all SNPs and the second set ( n =2203 cases and 2280 controls) is then tested for those SNPs that show marginally significant associations in set 1 ( P -heterogeneity or P -trend <0.1).
Described associations between PPARG polymorphisms and breast carcinoma are also discrepant: some researchers see a marginally significant increase in the risk of breast cancer among women homozygous for the Ala allele of PPAR γ (Pro12Ala), causing a reduction in the transcriptional activity of PPAR γ 2 [ 35 ], while others stress that carriers of the same variant allele are at lower risk [ 36 ].
Linkage disequilibrium measures are highly significant between SNPs rs1327328 and rs7326018 and SNP rs4284505, located next to a cluster of microRNAs, and marginally significant with SNP rs9583760, located within LOC121727 (similar to Peroxisome assembly protein 12 (Peroxin-12) (Peroxisome assembly factor 3) (PAF-3)).
To conclude that MT improved the behavior of the older sample of children, that it provided a ‘marginally significant’ increase in IQ, or that it ‘appears to be a viable therapy for promoting greater daily functioning and communication’ based on within-group analyses is misleading, because any or all of those pre–post effects could be observed as a result of the aforementioned threats even if MT was wholly ineffective.
