The resulting signature, 4-GES (consisting of SOCS2 , IL2RA , NPDC1 , and PHGDH ) was confirmed as an independent prognostic parameter in five of six additional data sets, including some that contained cytogenetically heterogeneous patient groups and/or employed different gene expression profiling technologies, and just fell short of significance in the sixth data set.
Analysis of the primary outcome measure revealed that supplementation with NEM® produced an absolute rate of response that was significantly better (ranging from 10.3% to 26.6% improvement) than placebo at all time points for both pain and stiffness, but fell short of significance for function and overall WOMAC, despite improving by 8.8% to 15.5% (see Table 2 ).
The main effect of group fell short of significance, though there was a non-significant trend for a shallower slope in the child SLI group (mean slope = 0.14, SD = 0.27) than in the other three groups: mean for child Typical = 0.38, SD = 0. 35; mean for adult SLI = 0.42, SD = 0.39; mean for adult Typical = 0.24, SD = 0.25; F (3, 55) = 2.21, p = .098.
In the course of one year (month 12) of intervention implementation, in a linear mixed model, there were changes from baseline in haemoglobin levels (Hb) among participants in the intervention arm compared to those in control arm (0.31 g/dL, 95% CI: 0.2–0.4, p < 0.001), this was on individual participant but fell short of significance on cluster adjustment ( Supplementary Table S3 ).
