Over the 4 study years, we noted a possible trend of decreasing utilization of ICI monotherapy, which could be attributable to decreased patient numbers in 2020 or secondary to an increase at our academic center in ICI-based clinical trial options that enrolled immunotherapy-naïve patients, who were thus excluded from this observational study.
Inhibition of ATR suggested a possible trend to increased p21 mRNA expression ( Figure 3 H). 3.4. siRNA Knockdown of DDR Kinases To confirm these pharmacological data, we employed a gene-silencing approach to individually knock down ATM, ATR and DNA-PK. siRNA inhibition caused selective and specific reduction of each target mRNA within 24 h and was maintained for at least 96 h ( Supplementary Figure S3 ).
18 , 22 Subgroup analyses of those with chronic hypertension suggest a possible trend toward small for gestational age, birth weight <10th centile (13.9% versus 19.7%; adjusted odds ratio 0.66, 95%CI 0.44‐1.00); however, it is notable that in this subgroup the primary perinatal outcome was no different (odds ratio 1.08, 95%CI 0.78‐1.51).
Indeed, the halogen bioisostere‐substituted analogs, such as 3 e (4‐Me) and 3 f (4‐CF 3 ), also exhibited enhanced activities, suggesting a more important role of the hydrophobic over the halogen bond effect in this context. Similarly, evaluation of the 3‐halogen substituted compounds ( 4 a – 4 d ) also indicated a possible trend of classic halogen bonding (3‐F ( 4 a )>3‐Cl ( 4 b )>3‐Br ( 4 c )≈3‐I ( 4 d ), Table 2 ), albeit with slightly weaker potency when compared with that of their 4‐substituted counterparts.
