Despite the relatively small sample size, one SNP was identified with a very significant P -value ( P = 5.5*10 -8 ) and several others were identified that were suggestive of significance ( P < 1.0*10 -6 ).

Results were considered genome-wide significant at p < 5 × 10 −8 , and suggestive of significance at p < 5 × 10 −6 .

Many additional candidate genes for FA exist based on diagnostic criteria that are difficult to differentiate from sensitization to food allergens, or have reached levels suggestive of significance ( p < 1 × 10 −5 ) but may have been impacted by sample size or other methodological limitations.

Of these 27 loci, five loci were identified as either genome-wide significant or suggestive of significance ( p < 1 × 10 −5 ) in a separate, previously published GWAS of one of the five traits. 118 loci were genome-wide significant for neuroticism, with 38 loci not identified in the univariate item-level GWASs ( Supplementary Table 6 ; Figure 1b , Figure 2b ).

Figure 1 presents the corresponding Manhattan plot showing the p -value (−log10 p ) of each SNP analyzed as well as indicating both thresholds for significance (the GWAS p -value of significance and the p -value suggestive of significance; p < 5 × 10 −5 ).

In total, 24 SNPs suggestive of significance ( p ≤ 10 −4 ) were identified for Alentejana cattle and 20 SNPs were identified for Mertolenga cattle.

A weaker association was identified in LG7, where 15 SNPs exceeded a threshold suggestive of significance ( p < 0.0001) (the location of the SNPs highly associated with sex that exceed the thresholds are shown in Supplementary Data 2 ).

The CYP2D6 gene was significantly associated with plasma concentrations of endoxifen ( P = 0.0008) and suggestive of significance for 4‐OH‐Tam ( P = 0.0074); individuals with higher activity scores had increased circulating endoxifen (Figure 2 a) and 4‐OH‐Tam (Figure 2 b) levels.

A two-sided p value < 0.05 in UVMR was considered suggestive of significance when p < 0.0167 (Bonferroni correction p = 0.05/3 exposures) was considered statistically significant.

Associations with a p < 0.05 were considered suggestive of significance, and associations with a p < 0.017 (Bonferroni correction, p = 0.05/3) were deemed statistically significant [ 20 ].

PFAS and birth outcomes: mediation by epigenetics Across all families with passing total DNA methylation data ( n = 141), there were several relationships suggestive of significance ( p < 0.1) between birth outcomes (gestational age at birth, or Fenton z-scores) and PFAS exposures, when controlling for the necessary confounders for statistical mediation analyses (Table 6 ).

Binomial logistic regression was done to identify the independent predictors of gestational diabetes mellitus outcomes in two steps—univariate analysis (step 1) followed by multivariate analysis using independent variables that were found to be significant ( P < 0.05) or suggestive of significance ( P < 0.100) in step 1.

Objective A recent high-density fine-mapping (ImmunoChip) study of genetic associations in rheumatoid arthritis (RA) identified 14 risk loci with validated genome-wide significance, as well as a number of loci showing associations suggestive of significance ( P = 5 × 10 −5 < 5 × 10 −8 ), but these have yet to be replicated.

A q -value of 0.2 was used to identify CpG sites that are suggestive of significance, and a stricter p -value cutoff of 9E-8 was used to call highly significant differentially methylated CpG sites ( Mansell et al., 2019 ).

All identified SNPs that reached significance or seemed to be suggestive of significance were visualised using Haploview software [55] .
