Even though sample sizes in the East Asian cohorts are small, we observed that 62% of the signals have supportive evidence in East Asian ancestry-only analysis, with the same direction of effect, and 20% of these signals are also nominally significant (binomial test p = 2.27 × 10 −5 , 95% confidence interval [CI] = 7%–100%) ( STAR Methods ).
14 , a SNP at the PIDD1 gene locus reached suggestive significance 14 ( P = 7 × 10 −7 ), and our lead novel variants at SSBP3 and THSD7A were at least nominally significant ( P = 2.61 × 10 −5 and P = 0.025, respectively).
Association analysis in Hanoverian stallions In 216 Hanoverian stallions, a nominally significant association with de-regressed EBV-PAT was found for the NOTCH1 variant g.37455302G>A ( P = 0.00003) using a model fitting the genotypes.
Among the 13 genes in which LGD strict variants were detected, only RET displayed nominally significant enrichment for rare damaging variants and passed the burden test ( P = 3.35E-05, FDR = 0.00047, other data not shown).
Nominally significant associations with PD risk were identified for four SNPs: XRCC6 rs5751129 ( p = 3.55 × 10−5), XRCC4 rs28360071 ( p = 0.0181), NBN rs2735383 ( p = 0.0213) and RAD50 rs17772583 ( p = 0.0411).
It is also nominally significant in the IGAP GWAS ( p -value = 3.86 × 10 −5 ).
Last, we evaluated the evidence for 14 previously published aneurysm risk single‐nucleotide polymorphisms through collaboration in extended aneurysm cohorts, with a total of 6548 cases and 16 843 controls (IA) and 4391 cases and 37 904 controls (AAA), and found nominally significant associations for IA risk locus 18q11 near RBBP8 to AAA (odds ratio [OR]=1.11; P =4.1×10 −5 ) and for TAA risk locus 15q21 near FBN1 to AAA (OR=1.07; P =1.1×10 −3 ).
Gene set enrichment analyses demonstrated nominally significant enrichment in the set related to collagen type XI trimer ( P = 4.18 × 10 −5 ) (Table S7 ).
This difference was also evident in the AFR populations from the UK Biobank and AASK, where both variants are common and show moderate, nominally significant effects after adjusting for G1/G2 ( p = 4.4 × 10 −5 and 4.5 × 10 −5 , respectively, Figure 4 B; Table 2 ).
Variants at PHLPP1 were also nominally significant in the independent meta‐analysis of African individuals that included WGS data ( p = 4.56 × 10 ‐5 ).
A recent GWA meta-analysis in Crohn's disease identified 74 independent SNPs as nominally significant ( p <5×10 −5 ) [24] .
From this meta-analysis, we ranked the SNPs according to their p -values and selected all the nominally significant SNPs with p -values < 5 × 10 −5 .
However, a number of methylation probes were nominally significant in each analysis ( p < 5 × 10 −5 , detailed in the Supplementary Information ).
When GWAS was performed in the 113 11q-deletion cases and 282 controls (78 undeleted 11q and 204 MNA neuroblastomas, Methods) of European-American ancestry, the 11q22.2 locus was still nominally significant (rs10895322, P = 5.50 × 10 −5 , OR = 2.811, logistic regression test, Supplementary Table 6 ), indicating a unique and independent role of the 11q22.2 locus in the 11q deletion cases.
Compared to results from the primary epigenome-wide mediation models, the mediation effects of CpGs across all cardiometabolic outcomes were slightly attenuated after accounting for smoking; however, all CpGs remained nominally significant (p < 5.6E-05).
Some of the CpG sites with nominally significant (p<6.7×10 −5 ) differences in methylation between HA and LA are close to genes that are differentially expressed in response to hypoxia; these genes include: toll-like receptor 6 ( TLR6) [57] , mif two 3 homolog 1 ( SUMO1) [27] ; phosphodiesterase 4A ( PDE4A) [58] and human immunodeficiency virus type I enhancer binding protein 2 ( HIVEP2 ) [42] .
A 2-tailed P < 0.05 for validation tests with FG and FI was considered nominally significant, and after the Bonferroni correction for multiple comparisons, P < 7E-05 for FG and P < 5E-04 for FI were considered statistically significant.
For EMBP, a downregulation in plasma was observed for both inhibitors (Figure 2 ) but was nominally significant for reslizumab ( p value = 7.16E‐05).
Finally, we verified that 13 out of the 16 life-shortening alleles are depleted in older study participants of the UK Biobank ( P =0.0021) and five of them have nominally significant association with participant age ( P =8.09 × 10 −5 ).
Also, although the association between rs174548 and FADS1 was nominally significant in tissues such as whole blood and subcutaneous adipose tissue, the associations did not reach the multiple testing correction threshold (FDR<0.05, p≤ 8.30x10 -5 ).
Nominally significant decreased DCM risk was found to be associated with the A allele and AT genotype of rs2505568 (OR: 0.48, 95% CI = 0.35–0.67, p < 0.0001 and OR: 0.44, 95% CI = 0.31–0.62, p < 0.0001, respectively), but it should be interpreted with caution because of Hardy-Weinberg disequilibrium in the control group.
rs35346340 is a significant FURIN eQTL in esophageal mucosa ( P = 2.0E-13) and a nominally significant eQTL of this gene in aortic artery ( P = 1.0E-04), and FURIN is highly expressed in both primary and immortalized cardiac fibroblasts.
