These results raise an extremely significant question about gene research and characterization: whether gene or its individual transcripts are used as basic functional units for gene research, characterization and application, such as gene annotation, pathway construction, functional categorization, gene editing, and genetic engineering.
In M1 macrophages, due to stimulation with LPS, the expression levels of these inflammatory cytokines increased sharply to 1871.08±147.20 pg/mL, 639.86±20.94 pg/mL, 778.19±28.30 pg/mL, 8698.74 pg/mL±395.70 pg/mL, 1754.30±107.87 pg/mL, and 1590.90±136.65 pg/mL, respectively, and there were extremely significant differences compared with the corresponding expression levels in M0 macrophages.
BrCTL1, BrCTL2 and BrCTL4 were mainly responsible for this trend, reaching ~3500, ~450, and ~9000 TPM, respectively, with extremely significant FC values following LPS stimulation: BrCTL4 was the fourth most highly expressed DEG in LPS-treated specimens; BrCTL1 was ranked 13th in the list of top 30 DEGs ( Table 2 ); BrCTL2 was within the 300 most highly expressed mRNAs.
Meanwhile, the transcriptomic analysis of the glutathione metabolism pathway in beef samples revealed that, compared to Qinchuan cattle and Simmental cattle, Pingliang Red Cattle samples had eight genes with extremely significant upregulation in their relative expression levels, and these expression levels were positively correlated with the glutathione content in the beef ( Figure 8 b).
In the CK area, H 10 , H 20 , and AN have weak correlations (correlation coefficients: 0.380 and −0.423); H 20 and OP, AK, and OM also show the same regular (correlation coefficients: −0.474, −0.472, −0.469); the correlations between H 20 , H 30 and AN, OP, AK, and OM in CMI area are extremely significant (correlation coefficients: 0.690, 0.693, 0.637, and 0.664 and 0.638, 0.787, 0.742, and 0.823, respectively).
