Specifically, in humans, we observed a dominant occurrence of C-mode 1 and 2 (Kruskal–Wallis test, p < 0.001) while mice occurrence of sensory-oriented C-mode 3 and 4 was observed instead (Kruskal–Wallis test, p < 0.001), with macaques showing equiprobable (Kruskal–Wallis test, p = 0.25) C-mode occurrence (although a slight trend for a mouse-like profile was apparent).
While there is a slight trend towards reduced death in the WT+AP20187+OVAp group in Figure 1B, there is a clear significant difference between this group and the MAFIA+AP20187+OVAp, suggesting that treatment with drug has the biggest impact on MAFIA slices, in which phagocytes are susceptible to deletion, and any potential off-target impacts of the drug on negative selection are relatively minimal.
In alignment with our flow cytometry analysis ( Figure 5 D), no pronounced changes in normalized protein abundances were detected for markers of oligodendrocyte maturation or myelination ( Figure 5 F, top), but in half of the ABEmax-treated mice (n = 3) we observed a slight trend for increased abundance of various markers for astrocyte maturation (subset 1 in Figure 5 F).
