Heterozygous carriers of INHBE pLOF variants (1 in 587 individuals, combined frequency of 0.08%) had a favorable metabolic profile consisting of nominally significant associations with lower triglycerides ( P = 9.65 × 10 −4 , Beta = −0.13 SD), higher HDL cholesterol ( P = 0.01, Beta = 0.10 SD), decreased alanine aminotransferase ( P = 0.04, Beta = −0.08 SD) and lower fasting glucose ( P = 0.03, Beta = −0.17 SD).

Lastly, statistical analysis revealed a highly significant (MonteCarlo, P =0.00099) correlation between DEGs of normal ageing versus bcat-1 RNAi, suggesting a global functional relevance of BCAT-1 for the process of physiological ageing.

Despite the generally low levels of genetic differentiation, the populations exhibit a highly significant signal of isolation-by-distance (Mantel test r = 0.79, p < 0.001) which becomes especially apparent due to the large geographical extent of our sampling (Fig. 2d ).

By using linear mixed model analyses testing for genetic variation among accessions, and by estimating the impact of the TM treatment on biomass accumulation compared to the respective mock control, we detected a highly significant accession-by-treatment interaction ( P < 0.001), highlighting natural variation in the accumulation of biomass of the accessions in response to chronic ER stress.
