While none of the 1463 Reactome pathways were significantly enriched for DC differential expression (corrected p -value at 0.05), two pathways were significantly enriched for AC (Platelet sensitization by LDL and Response to metal ions), three for TI (Cytoprotection by HMOX1, Haem signalling and Response to metal ions), and one was significant for R (Aflatoxin activation and detoxification), and four narrowly missed statistical significance, including two interferon signalling pathways—gamma and alpha/beta (corrected p -value at 0.059, Supplementary Fig. 7B ).
The reduction in risk of a decline in eGFR associated with coffee consumption narrowly missed statistical significance in participants with a long duration of diabetes (≥15 years) versus those with a shorter duration, but was significantly larger in participants with poor glycemic control (baseline HbA 1c ≥8%) than in those with better glycemic control.
TSA crossed the monitoring boundary under the selected assumptions; however, because the conventional meta-analysis narrowly missed statistical significance and TSA is sensitive to assumptions regarding anticipated effect size and control event rate, this finding should be interpreted cautiously and considered exploratory ( Figure 10 ).
As a sensitivity analysis, we also fit a model containing all candidate predictors without prior selection (Supplementary Table S2); effect estimates were consistent with the main analysis in direction and magnitude, although frailty narrowly missed statistical significance in the full model (aOR: 0.72, 95% CI: 0.51 to 1.02).
