We found very strong evidence of association of multiple SNPs at TTC12 and ANKK1 in two different populations, EAs and AAs (minimal P=0.0007 in AAs and minimal P=0.00009 in EAs), and highly significant association of a single haplotype (set of markers) spanning TTC12 and ANKK1 to ND in the pooled sample (P=0. 0000001).

As expected, there was significant enrichment of pathways involved in ‘bacterial invasion of epithelial cells’, ‘tight junctions’, ‘cytokine-cytokine receptor interactions’ and ‘colorectal cancer’; however, we also observed a highly significant association with ‘apoptosis’ (p<10 −7 ) ( online supplementary figure 6 ).
Our work also provides a list of 28 genes that are highly significant (p<10 −7 ) in all the three M1 stimuli and could be used as a starting point to define an agreed global M1 signature, when validated with other M1-polarizing agents ( e . g .
The binding profiles are extremely similar ( Figure 2 A), with a highly significant correlation between the two data sets (r = 0.586, p < 1 × 10 −7 , signal across genes).
Among 85 tissue cores with clear tumor and benign epithelium regions randomly selected for analysis, the difference in staining was highly significant ( P = 1 × 10 −7 ).
3 Discussion The first significant finding derived from this study is the clear and highly significant correlation between TD and response, with a response rate of only 9.1% for patients with TD <205 Gy against 89.7% for those with TD ≥205 Gy, P <10 −7 .
Further analysis of the gene set revealed a highly significant enrichment (p= 1.0 −7 ) for the upstream regulator hepatocyte nuclear factor 4 alpha (HNF4α), which regulates almost 200 genes (~ 15% of genes) in the list of differentially expressed genes between the two groups post-treatment ( Supplementary Table 1B ).
