An interesting trend that was observed in primary human cells, iPSCs, and several mouse organs was that a few AAV variants, such as AAV4 (multiple cell types and tissues, except the lungs), AAV5, and AAV12 (in a few targets, but especially in the case of kidney in the hFRG mouse), were very efficient at cell entry, but were unable to express the transgene at the RNA level.
However, using the log rank test to generate Kaplan–Meier curves, an interesting trend toward significance was observed in disease outcome (OS and RFS), with patients with higher SBEM levels being associated with worse survival and a shorter time to progression ( P = 0.063, HR 0.461, 95% CI of ratio 0.117, 1.060; and P = 0.072, HR 0.522, 95% CI of ratio 0.172, 1.078, respectively; Figure 4E,F ).
