As expected, in our multiomics study, disease severity followed an increasing trend from SDC to UDC peaking at the pre-ACLF group, whose patients had higher rate of ascites and infections, higher serum bilirubin, INR, creatinine, ALT, leukocyte and neutrophil counts, and CRP, and lower hematocrit, mean arterial pressure, and serum levels of sodium and albumin (Supplementary Table 1 ).
On the other hand, there was an increasing trend toward increased LRO risk in patients who carried more CMRFs than in those without any CMRF (aHR/CI: 1.35/1.02–1.78, 1.48/1.11–1.97, and 1.53/1.15–2.05 for 1, 2, and ≥ 3 CMRFs, respectively) (Table 2 and Figure 2C ). 3.4.
In CSF, ctDNA dynamics appeared to stratify by response outcome: patients with complete response/partial response (CR/PR) at both first assessment (at week 6) and best intracranial response showed low baseline ctDNA levels with a declining trend, whereas those with stable disease (SD) at both time points demonstrated relatively high baseline levels or an increasing trend ( Figure 4 C).
